EXCITED-STATE PROTON TRANSFER IN NUCLEIC ACID BASES, NUCLEOSIDES, AND THEIR ANALOGUES: A MINI-REVIEW Dedicated to Professor David Shugar on his 95-th birthday

چکیده

Fluorescent nucleic acid base analogues and the corresponding nucleosides that reveal excited-state proton transfer, including phototautomerism, are briefly characterized, and their potential applications as fluorescent probes discussed. Among the most promising systems are formycins and some 8-azapurines as well as the corresponding nucleosides, like 8-azaxanthosine. Other, nonfluorescent systems exhibiting phototautomeric behavior are reviewed, with special emphasis on the G-C dimer.

Xanthine, xanthosine and its nucleotides: solution structures of neutral and ionic forms, and relevance to substrate properties in various enzyme systems and metabolic pathways.

The 6-oxopurine xanthine (Xan, neutral form 2,6-diketopurine) differs from the corresponding 6-oxopurines guanine (Gua) and hypoxanthine (Hyp) in that, at physiological pH, it consists of a approximately 1:1 equilibrium mixture of the neutral and monoanionic forms, the latter due to ionization of N(3)-H, in striking contrast to dissociation of the N(1)-H in both Gua and Hyp at higher pH. In xan...

Nucleoside phosphate analogues of biological interest, and their synthesis via aryl nucleoside H-phosphonates as intermediates.

This review presents a brief account of the chemistry and mechanistic aspects of aryl H-phosphonates, and selected applications of this class of compounds as intermediates in the synthesis of a wide range of biologically important analogues of nucleoside phosphates, and oligonucleotides, in which the phosphate moieties are replaced by other structurally related groups. The aryl nucleoside H-pho...

Polyphosphate nucleosides of purine analogues.

To test the hypothesis that carcinostatic purine and pyrimidine analogues exert their effects at the nucleotide level, we have undertaken the preparation and study of nucleotides, including coenzymes, which contain analogues in place of the natural bases. An earlier communication from this laboratory (1) described the enzymatic synthesis and isolation of 5’-nucleotides of a number of purine ana...

Ultrafast non-radiative decay of gas-phase nucleosides.

The ultrafast photo-physical properties of DNA are crucial in providing a stable basis for life. Although the DNA bases efficiently absorb ultraviolet (UV) radiation, this energy can be dissipated to the surrounding environment by the rapid conversion of electronic energy to vibrational energy within about a picosecond. The intrinsic nature of this internal conversion process has previously bee...
